Using 5 identical materials gives a 100% chance to craft that material's associated potion, according to the update announcement.
Material Targeting

After the Alchemy System Rework, every confirmed material can be linked to an associated potion. When you add that material to the cauldron, the chance of getting its associated potion increases.
Adding more copies of the same material increases the chance further. If you use 5 identical materials, the associated potion is guaranteed.
However, stacking one material only increases the chance of that specific potion. It does not increase the chance of every potion in the same rarity.

Legendary
Legendary materials are the most important items to track after the rework because they can be reused when the final potion has a lower rarity. If a Legendary material is tied to a powerful Legendary potion, stacking 5 copies may become the most reliable way to target that potion.
Epic
Epic materials bridge the gap between filler and premium. They stack well for mid-to-high potion targeting and are returned if the result is Rare or below.
Rare
Rare materials are the most likely to be consumed in regular brewing. Stack them only if you want their specific potion — otherwise use them as general filler.
Example
If you place 2 Legendary materials, 1 Epic material, 1 Rare material, and 1 Uncommon material, but the result is a Rare Potion, only the Rare and Uncommon materials are consumed. The Epic and Legendary materials are returned.
Most non-event alchemy materials are being adjusted so their rarity lines up better with the potion rarity they help produce. This section will be updated as soon as the new material rarities are confirmed in-game.
